Разнообразие высокомолекулярных субъединиц глютенина и оценка генетического сходства яровой мягкой пшеницы, созданной в различных селекционных учреждениях

Аннотация

Актуальность. Исследования полиморфизма глютенина помогут выделить наиболее ценные генотипы для дальнейших  скрещиваний  и получения  новых  перспективных селекционных линий пшеницы с повышенными показателями качества. Цель исследования – идентифицировать  субъединицы  глютенина  и определить  аллельные состояния локусов Glu-1 в образцах яровой мягкой пшеницы.

 

Материалы  и методы.  Объектом  исследования  являлись 54 российских  и 76 казахстанских  сортов  и линий яровой  мягкой  пшеницы  из  различных  селекционных учреждений. Электрофорез глютенинов проводили в системе концентрирующего и разделяющего полиакриламидного  геля  по  методике  Лэммли.  Идентификация субъединиц глютенина – по каталогу Payne и Lawrence.

 

Результаты и обсуждение. На основе изучения коллекции пшеницы, по локусу Glu-A1 идентифицировано два аллеля: b  и c,  с различными  частотами  встречаемости. Установлено,  что  встречаемость  аллеля  Glu-A1b  выше Glu-A1с во всех образцах пшеницы. Идентифицировано шесть аллелей: а, b, c, d, f и g локуса Glu-B1. Наибольшая встречаемость зафиксирована для аллеля с (7+9). Локус Glu-D1 представлен двумя идентифицированными аллелями: a и d, с разными частотами встречаемости. В пшенице  из  представленных  селекционных  учреждений чаще присутствовали комбинации субъединиц глютенина: 2*, 7+9, 5+10 (9 баллов); 2*, 7+9, 2+12 (7 баллов) и Null, 7, 5+10 (6 баллов). На основе статистических расчетов достоверно отличалась челябинская пшеница от тюменской (локусы  Glu-A1  и Glu-B1) и от  восточно-казахстанской (локусы Glu-A1 и Glu-D1).
